A Correlative Study of the Proliferation Status and the Expression of Proliferating Cell Nuclear Antigen(PCNA) and C-H-ras Oncogene in Gastric Adenocarcinomas

Abstract
The flow cytometric DNA analysis and the immunohisiochemical staining of proliferating cell nuclear antigen(P.CNA) have been reported to reflect the cellular activities and the proliferative rates of the progression of neoplasms.
To evaluate whether there exist correlations between the proliferative activity by flow cytometric DNA analysis and the expression of c-H-ras oncogene. the author attempted the flow cytometric DNA analysis for the cellular activities and
immunohistochemical staining for PCNA and c-H-ras p21 protein on 89 cases of paraffin embedded tissues(15 cases of early gastric cancer, 74 cases of advanced gastric adenocarcinoma and 15 cases of chronic peptic ulcer).

The results were summarized as follows:
1. All cases of chronic peptic ulcer showed diploidy pattern in flow cytometric analysis.
2. The aneuploidy pattern was found in 3 of 15 cases(20%)of early gastric carcinoma and 36 of 74 cases(48.6%) of advanced gastric carcinoma.
3. There were significant differences between control gorup and early. and advanced gastric carcinomas in G0+G1 phase. S phase and G2+M/G0+G1 DNA ratio(P<0.05).
4. There was a positive correlation between percentage of PCNA positive cells and S Phase of flow cytometric result(r=0.22).
5. The c-H-ras negative cases had more frequency in lymph node metastasis and higher PCNA positivity
